RecombinantHumanLR3Insulin-likeGrowthFactor-1LR3-IGF1TheInsulin-likegrowthfactors(IGFs)aremitogenicpolypeptidegrowthfactors,predominantlyproducedintheliver,thatstimulatetheproliferationandsurvivalofvariouscelltypes,includingmuscle,boneandcartilagetissueinvitro.IGFsbelongtotheInsulingenefamilyandaresimilarbystructureandfunctiontoinsulin,buthaveamuchhighergrowth-promotingactivity.IGF1expressionisregulatedbygrowthhormone,whileIGF2expressionisinfluencedbyplacentalactogen.BothIGF1andIGF2signalthroughthetyrosinekinasetypeIreceptor(IGFIR).MatureIGFsaregeneratedthroughproteolyticprocessingofinactiveprecursorproteins.RecombinanthumanIGF1isaglobularproteincontaining70aminoacidsand3intra-moleculardisulfidebonds.TheLR3IGF1hasasubstitutionofargininefortheglutamicacidatposition3ofhumanIGF1andanextensionof13aminoacidresidues.Thischangeofsequencecausesittohaveamuchlongerhalf-lifeandisspecificallydesignedandmanufacturedformammaliancellculturetosupportlarge-scalemanufacturingofrecombinantbiopharmaceuticals.
