Product DescriptionThis product is produced by immunizing mice with a synthetic peptide corresponding to C terminus of human histone H2A.x. Antibodies are purified by peptide affinity chromatography.
SpecificityThis antibody can detect endogenous levers of total histone H2A.x protein. It does not cross-react with other histones or non histone proteins.

Important NotesFor Western blotting, incubate membrane with diluted antibody in 5% nonfat milk, 1 x TBS, 0.1% Tween-20 at 4℃ with gentle shaking, overnight.

BackgroundThe nucleosome, made up of four core histone proteins (H2A, H2B, H3, and H4), is the primary building block of chromatin. Nucleosomes wrap and compact DNA into chromatin, limiting DNA accessibility to the cellular machineries. Featuring a main globular domain and a long N-terminal tail, H4 is involved with the structure of the nucleosomes of the ‘beads on a string’ structure. Histone proteins are highly post-translationally modified however Histone H4 is the most extensively modified of the five histones. It is an important protein in the emerging field of epigenetics, where its sequence variants and variable modification states are thought to play a role in the dynamic and long term regulation of genes.
Storage & StabilityThe antibody is delivered in lyophilized powder. Upon receipt, please centrifuge the antibody at 12,000 x g for 20 seconds, and then reconstitute it with the antibody stabilizer provided. The stabilizer contains PBS, 50% glycerol, and 0.01% sodium azide. Please store the antibody at -20°C. Avoid repeated freeze/thaw cycles. Antibody is stable for 12 months from date of reconstitution. Equilibrate the antibody to room temperature for 2 minutes and mix by gently pipetting before use.**Research purposes only.
