The mouse-derived antibody is purified with protein G-conjugated agarose followed by affinity chromatography on immunogen peptide. It specifically recognizes endogenous histone H2B protein, not cross-react with other histones.

This product is produced by immunizing mice with a synthetic peptide corresponding to N terminus of human histone H2B. Antibodies are purified by peptide affinity chromatography.

For Western blotting, incubate  membrane with diluted antibody in 5% nonfat milk, 1 x TBS, 0.1% Tween-20 for  two hours at room temperature with gentle shaking.

Use at an assay dependent concentration. Optimal  dilutions/concentrations should be determined by the end user.

A.Western blotting analysis on 30 μg of crude proteins from human breast MCF-7, rat liver BRL and mouse neuroblastoma N2a whole cell lysates using histone H2B mouse mAb (1:2000).

B.Western blotting analysis on 30 μg of crude proteins from HeLa whole cell lysates with (“+”) or without (“-“) treatment of nocodazole (100 ng/ml, 18hr) and calyculin A (100 nM, 1hr) using histone H2B mouse mAb (1:2000).The membrane was re-probed by anti-phospho-histone H2A.x (Ser139) mouse mAb.

The nucleosome, made up of four core histone proteins (H2A, H2B, H3, and H4), is the primary building block of chromatin. Nucleosomes wrap and compact DNA into chromatin, limiting DNA accessibility to the cellular machineries. Featuring a main globular domain and a long N-terminal tail, H4 is involved with the structure of the nucleosomes of the ‘beads on a string’ structure. Histone proteins are highly post-translationally modified however Histone H4 is the most extensively modified of the five histones. It is an important protein in the emerging field of epigenetics, where its sequence variants and variable modification states are thought to play a role in the dynamic and long term regulation of genes.

Storage & StabilityThe antibody is delivered in lyophilized powder. Upon receipt, please centrifuge the antibody at 12,000 x g for 20 seconds, and then reconstitute it with the antibody stabilizer provided. The stabilizer contains PBS, 50% glycerol, and 0.01% sodium azide. Please store the antibody at -20°C. Avoid repeated freeze/thaw cycles. Antibody is stable for 12 months from date of reconstitution. Equilibrate the antibody to room temperature for 2 minutes and mix by gently pipetting before use.
