Themonoclonalantibody14D4reactswithmouseC3aR,whichisamemberoftherhodopsinsuperfamilyof7-transmembraneGprotein-coupledreceptors,withamolecularweightofapproximately54kDa.ExpressionofC3aRhasbeendemonstratedonawidevarietyofimmunecells,includingmonocytes,macrophages,dendriticcells,neutrophils,basophils,eosinophils,mastcells,TlymphocytesandBlymphocytes.Inaddition,C3aRisfoundoncellsofthecentralnervoussystem,lungsandkidney.InthecourseofcomplementactivationC3aRfunctionsasthecellsurfacereceptorforC3a,whichistheC-terminal77aminoacidcleavageproductofC3.C3a,possessesbothanaphylatoxicandimmunoregulatoryproperties,suchassmoothmusclecontraction,histaminereleasefrommastcells,andenhancedvascularpermeABIlity.Inaddition,C3ainducesrespiratoryburstinneutrophilsandhaschemotacticpropertiesforeosinophilsandmastcells.Moreover,C3acausesreleaseofkeycytokinesfrommultiplecelltypes,includingIL-1β,TNF-α,IL-6andIL-8.AroleforC3aRhasbeenimplicatedinseveralmurinediseasemodels.ItwasshownthatC3aRinhibitionreducesneurodegenerationinexperimentallupus,whereasinamurinemodelofallergicairwaydisease,deletionoftheC3areceptorprotectsagainstthechangesinlungphysiologyseenafterallergenchallenge.Finally,itwasshownthatdeletionofC3aRisprotectiveinmyelinoligodendrocyteglycoprotein-inducedexperimentalautoimmuneencephalomyelitis.
