EGFwasoriginallydiscoveredincrudepreparationsofnervegrowthfactorpreparedfrommousesuBMAxillaryglandsasanactivitythatinducedearlyeyelidopening,incisoreruption,hairgrowthinhibition,andstuntingofgrowthwheninjectedintonewbornmice.EGFisprototypicofafamilyofgrowthfactorsthatarederivedfrommembrane-anchoredprecursors.AllmembersofthisfamilyarecharacterizedbythepresenceofatleastoneEGFstructuralunit(definedbythepresenceofaconserved6cysteinemotifthatformsthreedisulfidebonds)intheirextracellulardomain.EGFisinitiallysynthesizedasa130kDaprecursortransmembraneproteincontaining9EGFunits.ThematuresolubleEGFsequencecorrespondstotheEGFunitlocatedproximaltothetransmembranedomain.ThemembraneEGFprecursoriscapableofbindingtotheEGFreceptorandwasreportedtobeBIOLOGicallyactive.
