FactorIX(akaChristmasFactor)isasingle-chain,415aminoacidglycoproteinzymogen.FactorIXisactivatedbyeitherFactorXIaortheFactorVIIacomplex.FactorIXaconvertsFactorXtoFactorXaduringtheintrinsicpathwayofthecoagulationcascade.FactorIXisusedtotreatpatientswithhemophiliaB,anX-linkedbleedingdisorder.ThesensitivequantitativemeasurementoftotalhumanFactorIXantigeninplasmasamplesiseasilyperformedwiththis96wellstripformatELISAkit.FactorIXandIXawillbedetectedbytheassay.TheconcentrationofFactorIXinnormalhumanplasmaisabout5ug/ml.TheassaymeasurestotalhumanFactorIXinthe0.1-100ng/mlrange.SamplesgivinghumanFactorIXlevelsabove100ng/mshouldbedilutedinblockingbufferbeforeuse.A1:500to1:1,000dilutionforplasmaissuggestedforbestresults.HumanFactorIXwillbindtothecaptureantibodycoatedonthemicrotiterplate.Afterappropriatewashingsteps,anti-humanFactorIXprimaryantibodybindstothecapturedprotein.Excessprimaryantibodyiswashedawayandboundantibodyisreactedwiththesecondaryantibody.Followinganadditionalwashingstep,TMBsubstrateisusedforcolordevelopmentat450nm.ColordevelopmentisproportionaltotheconcentrationofFactorIXinthesamples.AstandardcalibrationcurveispreparedusingdilutionsofpurifiedFactorIXandismeasuredalongwiththetestsamples.AllreagentsandstandardsareprovidedintheseELISAkits.
