Proreninisaglycosylatedasparticproteasethatconsistsof2homologouslobesandistheprecursorofrenin.Reninactivatestherenin-angiotensinsystembycleavingangiotensinogen,producedbytheliver,toyieldangiotensinI,whichisfurtherconvertedintoangiotensinIIbyACE,theangiotensin-convertingenzymeprimarilywithinthecapillariesofthelungs.Ithasbeenreportedthatthelevelsofcirculatingprorenin(butnotrenin)areincreasedindiabeticsubjects.Plasmaandserumconcentrationsincreaseinseveralconditionssuchaspregnancy,progressivediabetesmellitus,diabetesmellituswithmicrovasculardisease,anddiabeticretinopathy.Thesensitivequantitativemeasurementofhumanproreninincellculturemediaorurinesamplesiseasilyperformedwiththis96wellstripformatELISAkit.Activereninwillnotbedetectedbytheassay.Thiskitdoesnotcrossreactwithmouseorratprorenin.Theassaymeasureshumanprorenininthe0.01-10ng/mlrange.Samplesgivinghumanproreninlevelsabove10ng/mlshouldbedilutedinblockingbufferbeforeuse.Humanproreninwillbindtothecaptureantibodycoatedonthemicrotiterplate.Afterappropriatewashingsteps,anti-humanproreninprimaryantibodybindstothecapturedprotein.ExcessprimaryantibodyiswashedawayandboundantibodyisreactedwiththesecondaryantibodyconjugatedtoHRP.Followinganadditionalwashingstep,TMBsubstrateisusedforcolordevelopmentat450nm.Colordevelopmentisproportionaltotheconcentrationofprorenininthesamples.Astandardcalibrationcurveispreparedusingdilutionsofpurifiedprorenininblockingbufferandismeasuredalongwiththetestsamples.AllreagentsandstandardsareprovidedintheseELISAkits.
ThestandardcurveforthiskitisgeneratedusingTBS/BSAandisbestsuitedformeasurementofproreninincellculturemediaorurine.ThiskitisalsoavailablewithastandardcurveinprorenindepletedhumanplasmaforresearchersmeasuringprorenininplasmaorserumundercatalognumberHPRENKT.
