Abstract
Theaccurateandsensitivedetectionofchloramphenicol(CAP)isparticularlyimperativetopublichealthandsafety.Here,wepresentanovelsensorforresidualCAPdetectionbasedoncompetitivesurface-enhancedRamanscattering(SERS)immunoassayandmagneticseparation.Inthisnanosensor,functionalizedAunanoparticles(AuNPs)werelabeledwiththeRamanreportermolecule(e.g.4,4′-dipyridyl).WiththeadditionoffreeCAP,acompetitiveimmunereactionwasinitiatedbetweenfreeCAPandaboveAuNPsforconjugatingwithCAPantibody-modifiedmagneticnanoparticles(MNPs).Insteadofthesolidsubstrate,theantibodyconjugated-magneticbeadswereusedassupportingmaterialsandseparationtoolsinthepresentsensor.Withtheaidofamagnet,themixturewasremovedfromthesupernatantforconcentrationeffects.ThiscausedobviouschangeofSERSsignalintensityobtainedfromsupernatant.TheSERSsignalswerecollectedfromthesupernatantdirectly,whichmadetheSERSmeasurementsmorestable,repeatableandreliable.TheproposedSERS-basedmagneticimmunosensorallowsustodetectCAPinafast,selectiveandsensitive(1.0 pg/mL)manneroverawideconcentrationrange(1–1×10
4 pg/mL).Inaddition,theseresultsdemonstratethatthisimmunosensorholdsgreatpotentialforthe
detectionofantibioticsinrealaquaticenvironment,whichiscrucialtoourlife.
