Tocompletetheassay,standardorsamplecontainingTNF-sR2inthesolublephaseisaddedtomicrotiterwellscoatedwithanti-receptorantibody.TheTNF-sR2instandardorsampleiscapturedbythisantibodyonthesolidphase,andunboundmaterialsarewashedaway.Biotinylatedanti-receptorantibodyisthenadded,followedbyareactionwithStreptavidinHRPconjugatetolabelthebiotinylatedprobewithenzyme.Afterwashing,TNF-sR2boundtothecaptureantibodyonthestationaryphaseisdetectedwiththeadditionofachromogenicsubstrate.Sampleswithabsorbancevaluesabovethestandardcurveshouldbedilutedbeforeassay. |
