Background:
Arginaseenzymesconvertargininetoornithinethroughhydrolysis.TwoknownisoformsofArginaseexist,ARG1andARG2.Thisenzymeisinvolvedintheregulationofavarietyofimmunologicalresponsesandisamajortargetinimmunotherapy.ARG1/2isoverexpressedinmyeloid-derivedsuppressorcells(MDSCs)andtumor-associatedmacrophages(TAMs).OverexpressionofARG1/2resultsindepletedlevelsofargininebothintracellularlyandextracellularly.Asargininelevelsaredepletedinthemicroenvironment,immunecellsarestarvedofthisaminoacidandthefunctionofkeyimmunologicalactivatorsbecomeimpaired;Tcellproliferationisinhibited,regulatoryTcellsbecomeactivatedandinhibitCD4+Tcells,andimmunosuppressantshaveincreasedlongevity.DepletedargininealsoresultsinthereleaseofreactivenitrogenspeciesandreactiveoxygenspeciesfromTAMsandMDSCs.ThesereactivespeciescauseTcellapoptosisandtheactivationandgrowthofantigenpresentingcells.

TheARG1InhibitorScreeningAssayKitisdesignedtomeasureARG1enzymeinhibition.Thekitcomesinaconvenientformat,withenoughreactionsolutionandenzymetoperformatotalof400reactions.TheARG1InhibitorScreeningAssayKitissimpletouse.Inhibitorandenzymeareaddedtoasamplecontainingthioargininesubstrate.Afteraroomtemperatureincubation,activityisdeterminedbymeasuringtheabsorptionofreactionproductatλ=410–415nm.
