Specificity:ThisantibodyfragmentisspecifictotheDNAi-motif(Kd=59nM),whichisafour-strandedDNAstructurepotentiallyinvolvedinDNAregulationandformedinthecytosine-richregions.iMabantibodystructurewasshowntobeabletorecognisevariousi-motifs(1G22,1A83,c-MYC;RET,VEGF)locatedindifferentregionsofDNA(telomeres,centromeres,promotersofproto-oncogenes)whichmighthavedistincnucleotidesequences.Atthesametime,iMabantibodyfragmentdoesnotbindtomanyotherstructureswhicharesimilartoi-motifs,suchasdifferentDNAG4quadruplexforms,dsDNA;hairpinDNA;miRNAormutanthumantelomereswithoutthei-motif(ZerAATietal.,2018).ThissuggeststhatiMabisani-motifstructure-specificantibody.

ApplicationNotes:iMabisahighlysensitiveandspecificantibodyfragmentdirectedagainstDNAi-motifsanditisrecommendedfortheidentificationoftheseDNAstructures.Itsspecificitytowardsvarioustypesofthei-motifwasconfirmedviaELISAandbio-layerinterferometry(BLI)whichshowedthatiMabdidnotbindtoanysimilarstructuresandmolecules(mostimportantly,itdidnotbindtovariousDNAG4quadruplexforms,suchaspromoterG4sc-MYC44,BCL245,VEGF,telomeric2JSM47,2GKUandaptamerG4TBA)(Zeraatietal.,2018).iMabwasdemonstratedtobeeffectiveintheimmunofluorescencedetectionofi-motifsinmultiplecelllines(MCF7,U2OSandHeLa)simultaneouslyconfirmingthepresenceofi-motifstructuresinhumannucleiandtheirpHandcell-cyclestagedependence(Zeraatietal.,2018).
