Specificity:F124wasshowntoreactwithanepitopewithinthepre-S2regionoftheHBVsurfaceantigenMprotein,shownbyWesternblotandRIAusingbothfullviralparticlesandHBsAgparticles(Budkowskaetal,1986).F124isalsoreactiveagainstasyntheticpeptidecorrespondingtotheN-terminalresidues120-150(FPAGGSSSGTVNPVLTTASPL)ofthepre-S2region(Nietal,2010).HepatitisBisoneofthemostcommoninfectiousdiseasesonaglobalscale,withinfectionassociatedwithbothacuteandchronicliverinflammationandimplicatedinuptohalfofalloccurringhepatocellularcarcinomas.
ApplicationNotes:F124wasusedasasolid-phasecoatingforRIAsdetectingHBVparticles,resultinginparticleimmobilisationbybindingthepre-S2epitopeofthesurfaceantigen(Budkowskaetal,1986).AnscFv-formattedversionofF124,comprisingthesameVHandVLsequences,showeddose-dependentbindingto,andcompetitiveblockingofIgGandFabagainst,HBsAg(Passafiumeetal,1998).
