EGFwasoriginallydiscoveredincrudepreparationsofnervegrowthfactorpreparedfrommousesubmaxillaryglandsasanactivitythatinducedearlyeyelidopening,incisoreruption,hairgrowthinhibition,andstuntingofgrowthwheninjectedintonewbornmice.HumanEGFwasisolatedfromurinebasedonitsinhibitoryeffectongastricsecretionandnamedurogastrone,accordingly.EGFisprototypicofafamilyofgrowthfactorsthatarederivedfrommembrane-anchoredprecursors.AllmembersofthisfamilyarecharacterizedbythepresenceofatleastoneEGFstructuralunit(definedbythepresenceofaconserved6cysteinemotifthatformsthreedisulfidebonds)intheirextracellulardomain.EGFisinitiallysynthesizedasa130kDaprecursortransmembraneproteincontaining9EGFunits.ThematuresolubleEGFsequencecorrespondstotheEGFunitlocatedproximaltothetransmembranedomain.ThemembraneEGFprecursoriscapableofbindingtotheEGFreceptorandwasreportedtobebiologicallyactive.
AwidevarietyofbiologicaleffectshavebeenascribedtoEGFandothermembersoftheEGFfamily.ThebestdocumentedactivityofEGFisitsabilitytopromoteproliferationanddifferentiationofmesenchymalandepithelialcells.EGFisamitogenforfibroblasts,epithelialandendothelialcells,andpromotescolonyformationofepidermalcellsinculture.EGFinducesepithelialdevelopment,promotesangiogenesis,andinhibitsgastricacidsecretion.
