Signaltransducerandactivatoroftranscription(STAT),namedaftertheirdualrole,arelatentcytoplasmictranscriptionfactorsthatmediatevariousbiologicalresponses.TheactivationofSTATproteinsislargelymediatedbyphosphorylationofC-terminaltransactivationdomainthroughJanuskinases(JAKs)andmitogen-activatedproteinkinases(MAPKs),whichallowstheactivatedSTATstodimerizeandtotranslocateintothenucleus.Bymodulatingtargetgeneexpression,STATproteinsplayanimportantroleinmediatingabroadrangeofbiologicalprocessessuchascellproliferation,survival,apoptosis,anddifferentiation.SevenmammalianmembersoftheSTATfamilyareknown(STAT1,2,3,4,5a,5b,and6)andtheyallsharecommonfeaturesandstructure.STAT3isactivatedbygrowthfactorsandoncogenickinaseswhereitmediatestranscriptionalactivationofgenesencodingapoptosisinhibitors,cell-cycleregulatorsandinducersofangiogenesis.Transcriptionfactorthatbindstotheinterleukin-6(IL-6)-responsiveelementsidentifiedinthepromotersofvariousacute-phaseproteingenes.ActivatedbyIL31throughIL31RA.
