Quizartinib(AC220)isasecond-generationFLT3inhibitorthathasshownpromisingactivityinAMLinPhaseIIclinicaltrials.Itinhibitsmutantandwild-typeFLT3invivoat0.1and0.5µM,respectively,andhasshownfavorableactivityandtolerabilityinphaseIandIItrialsinacutemyeloidleukemia,withQTprolongationasthedose-limitingtoxicity.Recently,AC220hasalsobeenshowntobeaneffectiveinhibitorofATP-bindingcassette(ABC)proteinsABCB1(P-glycoprotein)andABCG2(breastcancerresistanceprotein).QuizartinibinhibitedtransportoffluorescentABCG2andABCB1substratesinABCG2-andABCB1-overexpressingcellsinaconcentration-dependentmanner,from0.1to5µMandfrom0.5to10µM,respectively,andinhibited[125I]-IAAPphotolabelingofABCG2andABCB1withIC(50)valuesof0.07and3.3µM,respectively.
