UnmethylatedCGdinucleotideswithinparticularsequencecontextsareresponsiblefortheimmunostimulatoryactivityofbacterialDNA.Syntheticoligonucleotides(ODN)thatcontainsuchCpGmotifs(CpGODNs)mimicmicrobialDNA.TheinnateimmunesystemofvertebrateshastheabilitytorecognizeCpGmotifsinmicrobialDNAviatheToll-likereceptor(TLR)9iftheCpGODNwerefreeofadditionalimmunestimulatorycontaminantsoftenpresentinsyntheticcommercialCpGODNpreparationsdesignedformolecularbiologyapplications(i.e.PCR).GiventhathighqualityCpGODNswereused[i.e.endotoxin-free],acloselinkhasbeenestablishedbetweentheexpressionofTLR9oncertainimmunecellsubsetsandthemodulationoftheimmunesystembyCpGDNA.DifferenttypesofCpGODNswereidentifiedbasedontheirdifferingbiologicaleffectsondifferentcelltypes:ODNTypeAisapotentinducerofIFN-αinhumanPDC,(i.e.ODN1585or2216)leadingtoantigenpresentingcell(APC)maturation,whereasODNTypeB(i.e.ODN2006orODN1668/ODN1826)isaweakinducerofIFN-αbutratherstimulatesIL-8productionandincreasingcostimulatoryandAg-presentingmoleculesandtriggersproliferationofB-cellsandIgMandIL-6production.AthirdtypeofCpGODNhasbeenidentified,termedODNTypeC,withbothhighinductionofINF-αinPDCandactivationofB-cells.ThesequenceofCpGTypeC(alsocalledK)(i.e.ODN2395orM362)combineselementsofbothTypeAandTypeBandcontainsacentralpalindromicsequencewithCGdinucleotides,acharacteristicfeatureofTypeA,andaTCGTCGmotifatthe5end,presentinTypeBCpGODNs.AlthoughtheCpGmotifsarethoughttodifferbetweenmiceandhumans,inbothspeciestherecognitionofCpGODNsismediatedbyTLR9.TheoptimalCpGmotifinhumansisGTCGTTandGACGTTforthemurinesequence.However,recentevidencesuggeststhatthissequencespecificityisrestrictedtophosphorothioate(PS)-modifiedODNandisnotobservedwhenanaturalphosphodiesterbackboneisused.Inrecentyearssequencerequirements,specificity,signalingpathwaysandkineticsoftheTLR9suppressionbyinhibitoryODNs(iODNs)havebeeninvestigated.
