MycoplasmasarespecificallydetectedbyamplifyingahighlyconservedrRNAoperon,ormoreprecisely,the16SrRNAcodingregioninthemycoplasmagenome.Themycoplasma-specificamplificationisdetectedat520nm(FAM™channel).The qOneStepPCRreactionmix includesprimersetsandFAM™labeledprobeswhichallowthespecificdetectionofallMollicutespeciessofardescribedascontaminantsofcellculturesandmediacomponents.EukaryoticDNAisnotamplifiedbythisprimer/probesystem.FalsenegativeresultsduetoPCRinhibitorsorimproperDNAextractionaredetectedbytheinternalamplificationcontrolwhich isalready includedinqOneStepreactionmix.Theamplificationoftheinternalamplificationcontrolisdetectedat610nm(HEX™channel).
