Brain-derivedneurotrophicfactor(BDNF)isaprosurvivalfactorinducedbycorticalneuronsthatisnecessaryforsurvivalofstriatalneuronsinthebrain.Itisasecretedproteinwiththemolecularweightof27.8kDa,consistingof247aminoacids.Itisknowntopromoteneuronalsurvivalanddifferentiation.BDNFsharessubstantialaminoacidsequenceidentitywithnervegrowthfactor(NGF).BDNFandneurotrophin-3(NT-3)aretworecentlyclonedneurotrophicfactorsthatarehomologoustoNGF.mRNAproductsoftheBDNFandNT-3genesaredetectedintheadulthumanbrain,suggestingthattheseproteinsareinvolvedinthemaintenanceoftheadultnervoussystem.BDNFandotherneurotrophinsarecriticallyinvolvedinlong-termpotentiation(LTP).BDNF-mediatedLTPisinducedpostsynaptically.BDNFhastrophiceffectsonSEROtonergic(5-HT)neuronsinthecentralnervoussystem.BDNFhasanessentialmaintenancefunctionintheregulationofanxiety-relatedbehaviorandinfoodintakethroughcentralmediatorsinboththebasalandfastedstate.Itplaysaroleintreatingbreathingdisorderssuchasrespiratoryinsufficiencyafterspinalinjury.ThematureformofBDNFisidenticalinallmammalsexamined,andthegeneencodinghumanBDNFtochromosome11,bandp13.
