Streptavidin is purified from the bacterium Streptomyces avidinii. It is a tetrameric protein that is composed of identical subunits, with the tetramer having a molecular weight of approximately 53kDa. Each subunit binds one biotin molecule with a very high degree of specificity and affinity. The Kd for this interaction is in the region of 10-15M.
Alkaline Phosphatase is a hydrolase enzyme. It is extracted from bovine intestine and purified using multiple precipitation, fractionation, chromatography, and filtration steps. Alkaline Phosphatase is frequently employed in immunoassays, and a commonly used colorimetric substrate for it is PNPP; the product of this reaction, p-nitrophenol, can be read at 405nm. Fluorogenic substrates such as 4-methylumbelliferyl phosphate (MUP) may also be used.
