Introduction

Endothelialcellsexpressthreedifferentvascularendothelialgrowthfactor(VEGF)receptors,belongingtothefamilyofreceptortyrosinekinases(RTKs).TheyarenamedVEGFR-1(Flt-1),VEGFR-2(KDR/Flk-1),VEGFR-3(Flt-4).Theirexpressionisalmostexclusivelyrestrictedtoendothelialcells,butVEGFR-1canalsobefoundonmonocytes,dendriticcellsandontrophoblastcells.Theflt-1genewasfirstdescribedin1990.Thereceptorcontainssevenimmunoglobulin-likeextracellulardomains,asingletransmembraneregionandanintracellularsplitedtyrosinekinasedomain.ComparedtoVEGFR-2theFlt-1receptorhasahigheraffinityforVEGFbutaweakersignalingactivity.VEGFR-1thusleadsnottoproliferationofendothelialcells,butmediatessignalsfordifferentiation.InterestinglyanaturallyoccuringsolublevariantofVEGFR-1(sVEGFR-1)wasfoundinHUVEsupernatantsin1996,whichisgeneratedbyalternativesplicingoftheflt-1mRNA.TheBIOLOGicalfunctionsofsVEGFR-1stillarenotclear,butitseemstobeanendogenousregulatorofangiogenesis,bindingVEGFwiththesameaffinityasthefull-lengthreceptor.

FLT1HumanRecombinantproducedinE.Coliissingle,anon-glycosylated,Polypeptidechaincontaining298aminoacidsfragment(31-328)correspondingtotheIgGlikedomains1-3fromthematuresolubleFLT1protein,havingatotalmolecularmassof43kDaandfusedwitha4.5kDaamino-terminalhexahistidinetag.
