Introduction

HepatitisBvirusisthemaincauseforhumanliverdisease,chronicinfectionfrequentlycauseslivercancerandcirrhosis.TheHBVcoregenecodes2distinctproteinproducts,a21.5-kDaproteinbeingassembledtoformnucleocapsidparticlesdesignatedHBcAg,whichwrapstheviralDNAaswellastheviralpolymeraseandRNaseH,andaprecoreprotein,designatedasHBeAg,whichisdirectlytotheendoplasmicreticulum,processedatN-andC-terminallyandsecretedasnon-particulatee-anitgen(HBeAg).Thepre-coreproteincontainsanextra29N-terminalaminoacids,servingasasignalpeptidetodirectthenascentpolypeptideintosecretorypathway.Afterthesecretion,matureHBeAgisdeletedattheresidue149C-terminallyandretains10precoreresiduesN-terminally.HBcAgandHBeAgaredistinctlyrecognizedbyantibodiesbuthighlycross-reactiveattheT-celllevel.
Theeantigenisfoundinthecirculation,itisfoundduringtheactiveHBVinfection,positiveresultindicatestheriskforcontagiousness,andisalsousedasindicatorfortheeffectivenessofHBVtreatment.Positiveanti-HBeAgspecifiesanactivestageofacuteHBVinfectionthatisinitsfinalstages,theriskforcontagiousnessisdramaticallyreduced.

RecombinanthepatitisBvirus“e”antigenproducedinE.coli,isa139a.a(10-149a.a)proteinandhavingamolecularmassofapproximately23kDa.TheHBeAgsequenceisderivedfromHBVadw2andfusedto6xHisTagatC-terminus.
