Background:

ThereactivityoftheantiserumisrestrictedtotheFcpartoftheIgMmolecule.InimmunoelectrophoresisandrADIalimmunodiffusion,usingvariousantiserumconcentrationsagainstnormalchickenserumasingleprecipitinlineisobtainedwhichshowsareactionofidentitywiththeprecipitinlineobtainedwithpurifiedIgM.NoprecipitationreactionisobtainedwithpurifiedIgG,IgA,andIgG/Fabfragments.InprecipitatingtechniquesasimmunoelectrophoresisandradialimmunodiffusiontoidentifythepresenceofIgMinchickenserumandotherbodyfluidsortodetermineitsconcentration.ToprepareanimmunoadsorbentforthepurificationofchickenIgMfromserumorplasma.

Precipitationassays.Inimmunoelectrophoresisuse2μlorequivalentagainst120μlantiserum.Indoubleradialimmunodiffusion(Ouchterlony)usearosettearrangementwith10μlina3mmdiametercenterwelland2μlserumsamples(neatandseriallydiluted)in2mmdiameterperipheralwells.

CrossReactivity:

TheantiSerumdoesnotcross-reactwithanyothercomponentoftheChickenIgsystem.Inter-speciescross-reactivityisanormalfeatureofantibodiestoimmunoglobulins,sinceIgofdifferentspeciesfrequentlyshareantigenicdeterminants.CrossreactivityofthisantiSerumhasnotbeentestedindetail.

Storage:

Thelyophilizedantiserumisshippedatambienttemperatureandmaybestoredat+4°C;prolongedstorageatorbelow-20°C.Dilutionsmaybepreparedbyaddingphosphatebufferedsaline(PBS,pH7.2).Repeatedthawingandfreezingshouldbeavoided.Ifaslightprecipitationoccursuponstorage,thisshouldberemovedbycentrifugation.Itwillnotaffecttheperformanceoftheantiserum.Dilutedantiserumshouldbestoredat+4°C,notrefrozen,andpreferablyusedthesameday.
