TheGenWayCandidaalbicansIgGAntibodyELISATestKithasbeendesignedforthethe detectionandthequantitativedeterminationofspecificIgGantibodiesagainstCandidaalbicansin serumandplasma.Furtherapplicationsinotherbodyfluidsarepossibleandcanberequestedfrom theTechnicalServiceofGenWay.
2.GeneralInformation
Invasivecandidalmycosis(synonyms:candidosis,candidiasis)accountformorethan80%ofthe casesofdeep-seatedmycosesinGermany.Thisincidenceissimilartothoseofotherindustrial countries.ThemaincausativeagentistheyeastCandidaalbicans,whichistheonlyfungalspecies belongingtothe„normal“microflora.Candida albicanshasalowpathogenicpotential,which rarelyallowsittoinfectahealthyhost.Rather,inimmunosuppressedindividualsitmaycause severaldiseasesasanopportunisticpathogen. Incandidalmycosisthreetypesofdiseasecanbedistinguished.First,Candidaalbicanscaninfect theskinandthemucousmembranesoftheouter genitals,mouth,respiratorytractanddigestion tract.Second,skinandmucousmembranesmaybeinfected,encouragedbyfactorslikepregnancy, diabetesmellitus,immunodeficiencyandtherapieswithcytostaticsorantibiotics.Third,the infestationoftheinnerorgans,forexamplethe lungsinaCandidasepsiscancausedeathin immunosuppressedindividualswithacellularimmunodeficiency. SerologicaltestsarefrequentlyusedtoascertaintheclinicalsignificanceofCandidaspp.isolates. RisingtitersoflgGantibodiestoCandidaalbicansmayreflectinvasivecandidiasisin immunocompetentindividuals.ThedetectionofIgAandIgMantibodiesisimportanttoidentifya freshinfection.Immunosuppressedindividualsoftenfailtoproduceantibodies,soanegative antibodytestdoesnotnecessarilyruleoutthedisease.Inthiscasetheuseofanantigendetection testisrecommended.Besidesthedetectionofantibodiesandantigen,thesiteofmanifestation,the numberofcolony-formingunits,andtheconcomitantuseofantimycoticsmustbeconsidered.
3.PrincipleoftheTest
TheGenWayCandidaalbicansIgGantibodytestkitisbasedontheprincipleoftheenzyme immunoassay(EIA).Candidaalbicansantigenisboundonthesurfaceofthemicrotiterstrips. Dilutedsampleserumorready-to-usestandardsarepipettedintothewellsofthemicrotiterplate.A bindingbetweentheIgGantibodiesoftheserum andtheimmobilizedCandidaalbicansantigen takesplace.Afteraonehourincubationatroomtemperature,theplateisrinsedwithdilutedwash solution,inordertoremoveunboundmaterial.Thenready-to-useanti-human-IgGperoxidase conjugateisaddedandincubatedfor30minutes.Afterafurtherwashingstep,thesubstrate(TMB) solutionispipettedandincubatedfor20minutes,inducingthedevelopmentofabluedyeinthe wells.Thecolordevelopmentisterminatedbytheadditionofastopsolution,whichchangesthe colorfrombluetoyellow.Theresultingdyeismeasuredspectrophotometricallyatthewavelength of450nm.TheconcentrationoftheIgGantibodiesisdirectlyproportionaltotheintensityofthe color.
