ThisproductcontainsTMB,substratebufferandhydrogenperoxideinasafe,readytousesolution.TheactivityofTMBhasbeenoptimisedtoenableincreasedsensitivity,minimalbackgroundandrapiddevelopment.
ThisproductproducesadeepbluecolourduringtheenzymaticdegradationofH2O2byhorseradishPeroxidase.Thereactionmaybestoppedwith0.2Msulphuricacid,resultinginayellowcolourreadat450nm.

1.Itisrecommendedthat100ulofGWB-Q00248TMBsubstrateisusedpermicrotiterwell.Pourthedesiredamountofsubstrateintoasealedcontainerandallowittoreachroomtermperatureinthedark.
2.Add100ulsubstratesolutionpermicrotiterwell.
3.Allowdevelopmentofthesubstratesolution.Timeofdevelopmentistypically5-30minutes.Forbestresults,theplateshouldbekeptinthedarkduringincubatione.g.wrappedintinfoil.
4.Forkineticassays,readabsorbanceat655nm(blue).Forendpointassays,addanequalvolumeof0.2Msulphuricacidandreadtheabsorbanceat450nm(yellow).Thisendpointsolutionisstableforuptoonehour.
