Short Description: In this assay the CRP present in serum sample reacts with the anti-TRX. antibodies which have been adsorbed to the surface of polystyrene microtitre wells. After the removal of unbound serum proteins by washing, anti- TRX. antibodies conjugated with horseradish peroxidase (HRP), are added. These enzymelabeled antibodies form complexes with the previously bound serum TRX. Following another washing step, the enzyme bound to the immunosorbent is assayed by the addition of a chromogenic substrate, 3,3’,5,5’-tetramethylbenzidine (TMB). The quantity of bound enzyme varies directly with the concentration of CRP in the sample tested; thus, the absorbance, at 450 nm, is a measure of the concentration of CRP in the test sample. The quantity of CRP in the test sample can be interpolated from the standard curve constructed from the standards, and corrected for sample dilution.

Reconstitution and Storage	1. DILUENT The 5X Diluent Concentrate is stable until the expira-tion date. The 1X working solution is stable for at least one week from the date of preparation. Both solutions should be stored at 4-8C. 2. WASH SOLUTION The 20X Wash Solution Concentrate is stable until the expira¬tion date. ENZYME-ANTIBODY CONJUGATE Undiluted horseradish peroxidase anti-CRP conjugate should be stored at 4-8C and diluted immediately prior to use. The reconstituted calibrator should be aliquoted out and stored frozen at least -20C (Avoid multiple freeze-thaw cycles). Kit Principle	Genway Biotech CRP ELISA Kit (Pig) (GWB-699A6F) is based on standard sandwich enzyme-linked immuno-sorbent assay technology. An antibody for C-reactive protein has been pre-coated onto a 96-wellplate (12 x 8 Well Strips). Standards or test samples are added to the wells, incubated and washed to remove unbound proteins. An Anti-C-reactive protein - HRP conjugated detector antibody is then added, incubated and unbound conjugate is washed away. An enzymatic reaction is produced through the addition of TMB substrate which is catalyzed by HRP generating a blue color product that changes to yellow after adding acidic stop solution. The density of yellow coloration read by absorbance at 450 nm is quantitatively proportional to the amount of sample C-reactive protein captured in the well.
