ShortDescription:InthisassaytheIgEpresentinsamplesreactswiththeanti-IgEantibodieswhichhavebeenadsorbedtothesurfaceofpolystyrenemicrotitrewells.Aftertheremovalofunboundproteinsbywashing,anti-IgEantibodiesconjugatedwithhorseradishperoxidase(HRP),areadded.Theseenzyme-labeledantibodiesformcomplexeswiththepreviouslyboundIgE.Followinganotherwashingstep,theenzymeboundtotheimmunosorbentisassayedbytheadditionofachromogenicsubstrate,3,3’,5,5’-tetramethylbenzidine(TMB).ThequantityofboundenzymevariesdirectlywiththeconcentrationofIgEinthesampletested;thus,theabsorbance,at450nm,isameasureoftheconcentrationofIgEinthetestsample.ThequantityofIgEinthetestsamplecanbeinterpolatedfromthestandardcurveconstructedfromthestandards,andcorrectedforsampledilution.
