Transmembraneserine/threoninekinaseactivintype-1receptorforminganactivinreceptorcomplexwithactivinreceptortype-2(ACVR2AorACVR2B).Transducestheactivinsignalfromthecellsurfacetothecytoplasmandisthusregulatingamanyphysiologicalandpathologicalprocessesincludingneuronaldifferentiationandneuronalsurvival,hairfollicledevelopmentandcycling,FSHproductionbythepituitarygland,woundhealing,extracellularmatrixproduction,immunosuppressionandcarcinogenesis.Activinisalsothoughttohaveaparacrineorautocrineroleinfolliculardevelopmentintheovary.Withinthereceptorcomplex,type-2receptors(ACVR2Aand/orACVR2B)actasaprimaryactivinreceptorswhereasthetype-1receptorslikeACVR1Bactasdownstreamtransducersofactivinsignals.Activinbindstotype-2receptorattheplasmamembraneandactivatesitsserine-threoninekinase.Theactivatedreceptortype-2thenphosphorylatesandactivatesthetype-1receptorsuchasACVR1B.Onceactivated,thetype-1receptorbindsandphosphorylatestheSMADproteinsSMAD2andSMAD3,onserineresiduesoftheC-terminaltail.Soonaftertheirassociationwiththeactivinreceptorandsubsequentphosphorylation,SMAD2andSMAD3arereleasedintothecytoplasmwheretheyinteractwiththecommonpartnerSMAD4.ThisSMADcomplextranslocatesintothenucleuswhereitmediatesactivin-inducedtranscription.InhibitorySMAD7,whichisrecruitedtoACVR1BthroughFKBP1A,canpreventtheassociationofSMAD2andSMAD3withtheactivinreceptorcomplex,therebyblockingtheactivinsignal.Activinsignaltransductionisalsoantagonizedbythebindingtothereceptorofinhibin-BviatheIGSF1inhibincoreceptor.ACVR1BalsophosphorylatesTDP2.
