Phospholamban(PLB/PLN)isasmalltransmembraneproteinwhichplaysanimportantroleincontrollingtheactivityofthesarcoplasmicreticulumATPase(SERCA2a)ofcardiacmuscleduringcalciumsequestration.
PhospholambanisphosphorylatedonseparateaminoacidresiduesbycAMP-dependent,andcGMP-dependent(Ser-16) andCa2+/CaM-dependent(threonine-17)proteinkinasesinresponsetoβ-adrenergicstimulation.Akthasalsobeenshowntophosphorylatethreonine-17.Theresultisanincreasedcalciumpumpactivitywhichreducesthetimecourseofthecalciumtransient,increasesthecalciumloadinthesarcoplasmicreticulum,andconsequently,producesalargercalciumtransientatthenextactionpotential.However,alterationinthishomeostaticinteractionhasbeenshowntoresultinheartfailure.
