TheRyanodineReceptorisaCa2+channelofcardiacmusclethatplaysacentralroleinECcoupling.ThebindingofCa2+toryanodinereceptor2(RyR2)opensthechannelandCa2+storedintheSRmovesthroughthechannelintothecytosoltoinitiatemusclecontraction.Abnormalstructureandfunctionofryanodinereceptorshasbeenreportedinfailinghearts,withSer-2808phosphorylationappearingelevatedinclinicalsituationswhichmaycontributetotheabnormalCa2+handlingcharacteristicsofcardiacmuscleintheseconditions.Ser-2808canbephosphorylatedinvitrobyPKAorCaMKII,whichiscoincidentwithsignificantchangeinRyR2channelfunctiontypifiedbyanincreasedopenprobABIlity,theabrogationoftheinhibitoryeffectsofCaMandMg2+,dissociationofregulatoryfactors,expressionofsubconductancestatesandtheexpressionofchannelactivityatdiastolicCa2+concentrations.Serine-2808inhumanRyR2equatestoSer-2809inrabbitRyR2.
