TheUCHT1antibodyisspecificforhumanCD3e,alsoknownasCD3epsilon,a20kDasubunitoftheTcellreceptorcomplex,alongwithCD3gammaandCD3delta.TheseintegralmembraneproteinchainsassemblewithadditionalchainsoftheTcellreceptor(TCR),aswellasCD3zetachain,toformtheTcellreceptor–CD3complex.Togetherwithco-receptorsCD4orCD8,thecomplexservestorecognizeantigensboundtoMHCmoleculesonantigen-presentingcells.TheseinteractionspromoteTcellreceptorsignaling(Tcellactivation),inducingcellproliferation,differentiation,productionofcytokinesoractivation-inducedcelldeath.CD3isdifferentiallyexpressedduringthymocyte-to-TcelldevelopmentandonallmatureTcells.
TheUCHT1antibodyisawidelyusedphenotypicmarkerforhumanTcells.Inaddition,binding/cross-linkingofUCHT1antibodytoCD3ecaninducecellactivation(useformatsuitableforfunctionalassays).ArecentpublicationofthecrystalstructureofaCD3e-antibodycomplexprovidesinsightastotheactionofcommonlyusedagonistantibodies,aswellasspecificepitope-bindingdataforthehumanCD3antibodiesUCHT1andOKT3(Fernandes,R.A.etal.2012.J.Biol.Chem.287:13324-13335).
UCHT1antibodyreactswithbothsurface-expressedandintracellularCD3eprotein,incontrasttoanalternativehumanCD3clone,Hit3a,whichwillstainonlytheextracellular(membrane-expressed)CD3eprotein.Also,theUCHT1antibodyisreportedtobecross-reactivewithchimpanzeeandhasbeenusedforphenotypicanalysisofexpressionbyflowcytometry;howevertheantibodyisreportedtobeunsuitableforinductionofTcellactivationinthisspecies(Bibollet-Rucheetal.2009.J.Virol.82:10271-10278).
