TheSK3antibodyreactswithhumanCD4,a59kDaproteinwhichactsasaco-receptorfortheTcellreceptor(TCR)initsinteractionwithMHCClassIImoleculesonantigen-presentingcells.TheextracellulardomainofCD4bindstothebeta-2domainofMHCClassII,whileitscytoplasmictailprovidesabindingsiteforthetyrosinekinaselck,facilitatingthesignalingcascadethatinitiatesTcellactivation.CD4,andco-receptorsCCR5andCXCR4,mayalsobeutilizedbyHIV-1toenterTcells.HumanCD4istypicallyexpressedonthymocytes,somematureTcellpopulationssuchasTh17andTregulatory(Treg)cells,aswellasondendriticcells.
TheSK3antibodyiswidelyusedasaphenotypicMarkerforhumanCD4expression,andhasbeenreportedtobecross-reactivewithRhesusandCynomolgusCD4.ThisantibodydoesnotblockbindingofalternativecloneRPA-T4,suggestingthattheyrecognizedifferentepitopes.
