TheGARP5antibodyreactswithhumanGARP,alsoknownasLRRC32orGarpin,an80kDatransmembraneproteinwhichactsasareceptorforthelatentformofTGF-beta1(pro-TGF-beta),preventingitssecretion.Specifically,GARPisreportedtoassociatewiththepro-domainofTGF-beta1,knownaslatency-associatedpeptide(LAP),whichmustbecleavedtoreleasethebiologicallyactivecytokine.TheputativeroleofGARPinsequesteringtheTGF-betaisimportantforregulatingitsactivity,asTGF-betasignalingisinvolvedindevelopmentandfunctionofTh17,Tregandmanyotherimmunecelltypes.GARPexpressionhasbeenshownonplateletsandathighlevelsonFoxp3+Tregcells,whereitisproposedtobeaphenotypicidentifierforactivatedhumanTregcells.
