TheCD28.2antibodyreactswithhumanCD28,a44kDatypeIsurfaceglycoproteinwhichactsasaco-stimulatoryreceptorinsupportoftheTcellreceptor(TCR).CD28existsasahomodimerwithspecificityfortwoknownligands,knownasB7-1(CD80)andB7-2(CD86),whichareexpressedonactivatedBcellsandantigen-presentingcells.TheseligandstriggerCD28signalinginconcertwithTCRactivationtodriveTcellproliferation,inducehigh-levelexpressionofIL-2,impartresistancetoapoptosis,andenhanceTcellcytotoxicity.Theinteraction/co-stimulatorysignalingbetweentheB7ligandsandCD28providescrucialcommunicationbetweenTcellsandBcellsorAPCstocoordinatetheadaptiveimmuneresponse.OthermembersoftheCD28familyofreceptorsincludeCTLA-4(CD152),PD-1(CD279),ICOSandBTLA.
TheCD28.2antibodymaybeusedasaphenotypicMarkerforhumanCD28,expressedonallCD4+TcellsandCD8+Tcells,andiswidelyusedasareagentforactivationoftheCD28receptorinvitroandinvivo(useformatsuitableforfunctionalassays).Thisantibodyisalsoreportedtobecross-reactivewithseveralnon-humanspecies,includingBaboon,Chimpanzee,Cynomolgus,andRhesus.
