TheRPA-T4antibodyreactswithhumanCD4,a59kDaproteinwhichactsasaco-receptorfortheTcellreceptor(TCR)initsinteractionwithMHCClassIImoleculesonantigen-presentingcells.TheextracellulardomainofCD4bindstothebeta-2domainofMHCClassII,whileitscytoplasmictailprovidesabindingsiteforthetyrosinekinaselck,facilitatingthesignalingcascadethatinitiatesTcellactivation.CD4,andco-receptorsCCR5andCXCR4,mayalsobeutilizedbyHIV-1toenterTcells.HumanCD4istypicallyexpressedonthymocytes,somematureTcellpopulationssuchasTh17andTregulatory(Treg)cells,aswellasondendriticcells.
TheRPA-T4antibodyiswidelyusedasaphenotypicMarkerforhumanCD4expression,andiscross-reactivewithChimpanzeeCD4.Thisantibodyrecognizesadifferentepitope,andthusdoesnotblockbindingof,thealternativeAnti-HumanCD4antibodycloneOKT4(ReinherzEL,etal.1979.Proc.Natl.Acad.Sci.76:4061-4065).
