ThePC61.5antibodyisspecificformouseCD25,a55kDasurfaceproteinalsoknownastheInterleukin-2Receptoralphachain,orIL-2Ralpha.CD25maybindIL-2byitself,althoughwithlowaffinityandwithoutinductionofcellsignaling.CD25isalsoexpressedwithinahigh-affinitycomplex,alongwiththeIL-2Rbetachain(CD122)andthecommongammachain(CD132),toformasignalingreceptorcomplex.ExpressionofCD25variesduringdevelopmentalstagesofTandBcells,isinducedonactivatedmatureTandBcells,andispresentonsubsetsofdendriticcells.CD25signalingaspartoftheIL-2receptorcomplextriggersTcellactivationandproliferation,aswellasmodulatingthedifferentiationandfunctionofTh17cells,Tregulatory(Treg)cells,anddendriticcells.
ThePC61.5antibodyisusedasaMarkerforTcells,Bcellsanddendriticcellsubsets.ExpressionofCD25,CD4andthetranscriptionfactorFoxp3isregardedasaphenotypicsignatureforTregcells.Assuch,thisantibodyiswidelyusedtodistinguishTregcellsfromnaïveorconventionalTcellswhichareCD25-.ThisclonehasalsobeenreportedfordepletionofTregcellsinvivo(useformatsuitableforfunctionalassays).
