TheTBhP3antibodyreactswithhumanFoxp3,a50-55kDatranscriptionfactorwhichisacentralregulatorofTcellactivityandiscriticalforthedevelopmentandfunctionofregulatoryTcells(Tregs).Foxp3,alsoknownasScurfin,JM2,orIPEX,isamemberoftheforkhead/winged-helixfamilyoftranscriptionalregulatorsandisaMarkerofbothnaturalandinducedTregs.MutationsinFoxP3areassociatedwiththeX-linkedIPEXsyndromeinhumansandwiththeX-linkedmutationinmicethatisresponsiblefor‘Scurfy’.Intracellularstainingofhumanperipheralbloodmononuclearcells(PBMCs)withTBhP3antibodyusingtheFoxp3/TranscriptionFactorStainingBufferKitrevealsapproximately0.5-4%oflymphocytesstaining,withthemajorityfoundintheCD25-brightpopulation.Thereissignificantvariationbetweendonors.
