TheN418antibodyreactswithmouseCD11c,alsoknownasintegrinalphaX.This150kDacellsurfaceglycoproteinispartofafamilyofintegrinreceptorsthatmediateadhesionbetweencells(cell-cell)andcomponentsoftheextracellularmatrix,e.g.fibrinogen(cell-matrix).Inaddition,integrinsareactivesignalingreceptorswhichrecruitleukocytestoinflammatorysitesandpromotecellactivation.Complete,functionalintegrinreceptorsconsistofdistinctcombinationsofintegrinchainswhicharedifferentiallyexpressed.IntegrinalphaX(CD11c)assembleswithIntegrinbeta-2(CD18)intoareceptorcomplexknownasCR4whichcanbindandinducesignalingthroughICAMsandVCAM-1onendothelialcellsandcanalsofacilitateremovalofiC3bbearingforeigncells.
TheN418antibodyiswidelyusedasaMarkerforCD11cexpressionondendriticcells(DC),ofteninparallelwithmarkersforCD11b,foridentificationofdevelopmentalstagesandmaturesubsetsofthiscelltype.CD11cisprominentlyexpressedontissuemacrophages,andisalsodetectedonsometypesofactivatedTcellsandintestinalintraepitheliallymphocytes(IEL).
