The3.9antibodyreactswithhumanCD11c,alsoknownasintegrinalphaX.This150kDacellsurfaceglycoproteinispartofafamilyofintegrinreceptorsthatmediateadhesionbetweencells(cell-cell)andcomponentsoftheextracellularmatrix,e.g.fibrinogen(cell-matrix).Inaddition,integrinsareactivesignalingreceptorswhichrecruitleukocytestoinflammatorysitesandpromotecellactivation.Complete,functionalintegrinreceptorsconsistofdistinctcombinationsofintegrinchainswhicharedifferentiallyexpressed.IntegrinalphaX(CD11c)assembleswithIntegrinbeta-2(CD18)intoareceptorknownasCR4whichcanbindandinducesignalingthroughICAMsandVCAM-1onendothelialcellsandcanalsofacilitateremovalofiC3bbearingforeigncells.
The3.9antibodyiswidelyusedasaMarkerforCD11cexpressionondendriticcells(DC),ofteninparallelwithmarkersforCD11b,foridentificationofdevelopmentalstagesandmaturesubsetsofthiscelltype.CD11cisprominentlyexpressedontissuemacrophages,andisalsodetectedonactivatedneutrophils,granulocytes,sometypesofactivatedTcellsandintestinalintraepitheliallymphocytes(IEL).Theantibodyisreportedtobecross-reactivewithBaboon,Chimpanzee,CynomolgusandRhesusCD11c.
