TrisomyX(TripleX)syndromeoccursatafrequencyofapproximately1in1000newbornfemales.Thesyndromeisassociatedwithanincreasedriskofseizuresandmayaffectshort-termmemoryandlearningalthoughthemajorityofwomenaffectedshowfewclinicalsymptoms.AxolhasdevelopediPSC-derivedneuralstemcellsfromapatientwiththetripleXchromosomalabnormality.AlloftheiPScellsexamined(20individualcellskaryotyped),hadtrisomyX.
AxolHumaniPSC-DerivedNeuralStemCells(NSCs)arederivedfromintegration-free,inducedpluripotentstemcells(iPSCs)underfullydefinedneuralinductionconditions.TheNSCsexpresstypicalMarkersofcerebralcorticalneuralstemandProgenitorcellssuchasPAX6,FOXG1andnestin,andspontaneouslyformpolarizedneuraltube-likerosettestructureswhenplatedasamonolayerinculture(seebelow).Additionally,AxolNSCsarecapableofgeneratingaspectrumofcerebralcorticalexcitatoryandinhibitoryneuronsthatareelectricallyactiveandhavetheABIlitytoformfunctionalsynapsesandcircuitsinvitro.Afterthawingandplating,theneuralstemcellsterminallydifferentiatetoacquirematureelectrophysiologicalproperties,andformfunctionalsynapticnetworksoveraperiodof40~50days.
AxolNSCsareeasytodifferentiatetoneuronsormixedneuralcelltypes,followingourprotocolsandusingourtailoredmediaandreagentbundles.AhighlypurepopulationofneuronscanbegeneratedfromAxolNSCsfollowingthesynchronousdifferentiationprotocol.Usingourspecializedcoatingreagents,neuronsderivedfromAxolNSCscanbemaintainedinculturelong-term(>1year).NSCsareavailablefrommultipledonorstosuityourresearchneedsandhavebeencharacterizedextensively.
