Cell Enrichment

Positive Selection

CD34⁺ cells are isolated from human bone marrow (BM) using an immunomagnetic cell separation techniques. These cells are positively selected using an antibody that targets the CD34 antigen.

 

Purity

The purity of CD34⁺ cells is >90% as measured by flow cytometric analysis.

 

Sample Collection and Processing

BM cells are drawn from the posterior iliac crest of normal human donors using a syringe containing anticoagulant.  MNCs are first obtained by density centrifugation of BM cells and CD34⁺ cells are then enriched from the MNCs by positive selection. BM cells are processed within 24-30 hours after collection.Donors have been screened to be negative for HIV, HBV, and HCV (see Certificate of Analysis).

 

Pre-Qualified for CFC Assays

These BM CD34⁺ cells are REACHBIO QUALIFIED™. They have been functionally pre-qualified in a colony-forming cell (CFC) assay using ReachBio’s ColonyGEL™ methylcellulose-based media. The frequency of myeloid and erythroid progenitors, as well as a recommended plating concentration for the CFC assay, are provided for each batch of these REACHBIO QUALIFIED™ cells (see Certificate of Analysis).

 

Stability and Storage

BM CD34⁺ cells are cryopreserved in CryoStor® CS5, a serum-free freezing medium.Cells are stable for long-term storage at -152^oC or in the gaseous phase of liquid nitrogen.Storage at -80^oC is not recommended. Once thawed, the cells must be used immediately and have a finite life span in culture.
