Humanlightdensityperipheralbloodmononuclearcells(PBMCs)areobtainedbydensitycentrifugation(Ficoll)ofaleukapheresisproduct.

SampleCollectionandProcessing

Peripheralbloodcellsarecollectedfromnormalhumanvolunteersvialeukapheresis(COBE®SpectraMNCcollectionprotocolorequivalent).Themononuclearcell-richleukocytelayer(leukapheresisproduct)iscontinuallyaspiratedbyalowflowratecollectionpumpdirectlyintoacollectionbag.MNCsarethenfurtherenrichedbydensitycentrifugation.
Peripheralbloodcellsareprocessedwithin24-30hoursofcollection.DonorshavebeenscreenedandfoundtobenegativeforastandardpanelofinfectiousagentsthatatminimumincludesHIV,HBV,andHCV.Dependingonthespecificlot,otherinfectiousagentsmayalsohavebeentestedfor(CertificateofAnalysisincludeslot-specifictestinginformation).

 

StABIlityandStorage

Peripheralbloodmononuclearcells(PBMCs)arecryopreservedinCryoStor®CS5,aserum-freefreezingmedium.Cellsarestableforlong-termstorageat-152oCorinthegaseousphaseofliquidnitrogen.Storageat-80oCisnotrecommended.Oncethawed,thecellsmustbeusedimmediatelyandhaveafinitelifespaninculture.
