CD8+CytotoxicTCellsareenrichedfromhumanperipheralblood(PB)mononuclearcells(MNCs)usinganegativeselectionimmunomagneticseparationmethodinvolvingofanantibodycocktailthattargetsanddepletesnon-CD8+cells.

 

SampleCollectionandProcessing

Peripheralbloodcellsarecollectedfromnormalhumanvolunteersvialeukapheresis(COBE®SpectraMNCcollectionprotocolorequivalent).Themononuclearcell-richleukocytelayer(leukapheresisproduct)iscontinuallyaspiratedbyalowflowratecollectionpumpdirectlyintoacollectionbag.ImmunomagneticcellenrichmentofCD8+cellsisperformedwithin30hoursofcollection.DonorshavebeenscreenedandfoundtobenegativeforastandardpanelofinfectiousagentsthatatminimumincludesHIV,HBVandHCV.Dependingonthespecificlot,otherinfectiousagentsmayalsohavebeentestedfor(CertificateofAnalysisincludeslot-specifictestinginformation).

PurityofCD8+CytotoxicTCellsis>90%asmeasuredbyflowcytometricanalysis.

 

StABIlityandStorage

PeripheralbloodCD8+cellsarecryopreservedinCryoStor®CS5,aserum-freefreezingmedium.Cellsarestableforlong-termstorageat-152oCorinthegaseousphaseofliquidnitrogen.Storageat-80oCisnotrecommended.Oncethawed,thecellsmustbeusedimmediatelyandhaveafinitelifespaninculture.
