Negative Selection

CD8⁺ Cytotoxic T Cells are enriched from human peripheral blood (PB) mononuclear cells (MNCs) using a negative selection immunomagnetic separation method involving of an antibody cocktail that targets and depletes non-CD8⁺ cells.

 

Sample Collection and Processing

Peripheral blood cells are collected from normal human volunteers via leukapheresis (COBE^® Spectra MNC collection protocol or equivalent). The mononuclear cell-rich leukocyte layer (leukapheresis product) is continually aspirated by a low flow rate collection pump directly into a collection bag. Immunomagnetic cell enrichment of CD8⁺ cells is performed within 30 hours of collection.Donors have been screened and found to be negative for a standard panel of infectious agents that at minimum includes HIV, HBV and HCV. Depending on the specific lot, other infectious agents may also have been tested for (Certificate of Analysis includes lot-specific testing information). 

Purity

Purity of CD8⁺ Cytotoxic T Cells is >90% as measured by flow cytometric analysis.

 

Stability and Storage

Peripheral blood CD8⁺ cells are cryopreserved in CryoStor® CS5, a serum-free freezing medium.Cells are stable for long-term storage at -152^oC or in the gaseous phase of liquid nitrogen.Storage at -80^oC is not recommended. Once thawed, the cells must be used immediately and have a finite life span in culture.
