Background

Thealpha-crystallinsaremajorwater-solublelensstructuralproteinsofthevertebrateeyethatarerelatedtothesmallheatshockproteinfamily.Thealpha-crystallinspossessstructuralandfunctionalsimilaritieswithHsp25andHsp27.Mammalianlenscystallinsaredividedintoalpha,betaandgammafamilies.Alphaandbetafamiliesarefurtherdividedintoacidicandbasicgroups(Alpha-AandAlpha-Brespectively).Inthelens,alpha-crystallinprimarilyfunctionstomaintainproperrefractiveindex,howeveritcanalsofunctionasamolecularchaperonethatbindstothedenaturedproteins,keepingtheminsolutionandtherebymaintainingthetranslucencyofthelens.Whencellularstressoccurs,alpha-crystallinentersits’phosphorylatedstateandmayserveastructuralcontrolfunctionandplayaroleinproteinmaintenance.Inadditiontotheirinteractionwithproteins,alpha-crystallinsalsointeractwithnativemoleculessuchasmembraneproteins,Golgimatrixprotein,structuralproteins,nuclearproteinsandDNA.Twootherfunctionsareanautokinaseactivityandparticipationintheintracellulararchitecture,andithasalsobeenproventhatbothalpha-AandBpreventapoptosisbyinhibitingcaspases.Specifically,alpha-Bcystallinisfoundinmanycellsandorgansoutsidethelens,andalphaBisoverexpressedinseveralneurologicaldisordersandincelllinesunderstressconditions.
