Background

Accumulationoftheamyloid-betapeptide(Abeta)inthecerebralcortexisacriticaleventinthepathogenesisofAlzheimer"sdisease.Thebeta-amyloidproteinprecursor(APP)iscleavedbyoneoftwobeta-secretases(BACEandBACE2),producingasolublederivativeoftheproteinandamembraneanchored99-aminoacidcarboxy-terminalfragment(C99).TheC99fragmentservesassubstrateforgamma-secretasetogeneratethe4kDaamyloid-betapeptide(Abeta),whichisdepositedintheAlzheimer"sdiseasepatient"sbrains.Recently,DeathReceptor6(DR6)wasfoundtointeractwithanamino-terminalfragmentofthebeta-amyloidprotein(N-APP)inneurons,activatingacaspase6-dependentapoptoticeventleADIngtoaxonaldegenerationandpruningduringdevelopment,suggestingthatthesetwoproteinsareinvolvedinneuraldevelopmentandmaypossIBLyplayaroleinAlzheimer"sdisease.
