AKTisacomponentofthePI-3kinasepathwayandisactivatedbyphosphorylationatSer473andThr308.AKTisacytoplasmicproteinalsoknownasAKT1,ProteinKinaseB(PKB)andrac(relatedtoAandCkinases).AKTisakeyregulatorofmanysignaltransductionpathways.AKTexhibitstightcontrolovercellproliferationandcellviability.OverexpressionorinappropriateactivationofAKTisnotedinmanytypesofcancer.AKTmediatesmanyofthedownstreameventsofPI3-kinase(alipidkinaseactivatedbygrowthfactors,cytokinesandinsulin).PI3-kinaserecruitsAKTtothemembrane,whereitisactivatedbyPDK1phosphorylation.Oncephosphorylated,AKTdissociatesfromthemembraneandphosphory-latestargetsinthecytoplasmandthecellnucleus.AKThastwomainroles:(i)inhibitionofapoptosis;(ii)promotionofproliferation.
