APH1wasinitiallyidentifiedasacomponentoftheNotchpathwayinC.elegans.Alongwithnicastrin,PEN2,andpresenilin-1APH1isanessentialcomponentofthegamma-secretasecomplexwhichcleavetheamyloidprecursorprotein(APP)atwhatareknownasthegamma-andepsilon-sitesandcanleadtotheaccumulationoftheAmyloidbetapeptide(Abeta)cleavageproductthatisassociatedwithAlzheimersdisease.APH1existsinatleastthreedistinctisoformswithAPH1aastheprincipalisoformpresentinthegamma-secretasecomplex.Micedeficientinthisisoform,butnottheothertwo,werelethalatE10.5,withimpairedvascularandneuraldevelopmentobserved.
