Background

Anti-AlphaInternexinantibodyrecognizesalpha-internexinwhichisaClassIVintermediatefilamentoriginallydiscoveredasitco-purifieswithotherneurofilamentsubunits.Alpha-internexinisrelatedtobutdistinctfromthebetterknownneurofilamenttripletproteins,NF-L,NF-MandNF-H,havingsimilarproteinsequencemotifsandasimilarintronorganization.Itisexpressedonlyinneuronsandinlargeamountsearlyinneuronaldevelopment,butisdown-regulatedinmanyneuronsasdevelopmentproceeds.Manyclassesofmatureneuronscontainalpha-internexininadditiontoNF-L,NF-MandNF-H.Insomematureneuronsalpha-internexinistheonlyneurofilamentsubunitexpressed.Antibodiestoalpha-internexinarethereforeuniqueprobestostudyandclassifyneuronaltypesandfollowtheirprocessesinsectionsandintissueculture.Inaddition,recentstudiesshowamarkedup-regulationofalpha-internexinduringneuronalregeneration.Theuseofantibodiestothisproteininthestudyofbraintumorshasnotbeenexaminedtodate,butislikelytobeofinterest.RecentlyCairnsetal.usedthisantibodytoshowthatalpha-internexinisanabundantcomponentoftheinclusionsofneurofilamentinclusionbodydisease(NFID),aserioushumanneurodegenerativedisorder.Theantibodywasalsousedtoconfirmthepresenceofcirculatingauto-antibodiestoalpha-internexinintheseraofsomepatientswithendocrineautoimmunity,aswellasinsomenormalindividuals.Anti-AlphaInternexinantibodyisidealforinvestigatorsinvolvedinCellSignaling,Neuroscience,SignalTransductionresearch.
