Background

Adhesiontoextracellularmatrixregulatescellsurvivalthroughbothintegrinengagementandappropriatecellspreading.Anoikisisthemolecularmechanismofapop-tosisinducedbyintegrindetachment.Amino-terminalenhancerofsplit(AES)isamemberoftheGroucho/transducin-likeenhancerofsplit(TLE)familyoftranscriptionalregulators,agroupoftranscriptionalco-repressorsthatplayimportantrolesinneurogenesis,segmentation,andsexdetermination.AESformsacomplexwithBit1(Bcl-2inhibitoroftranscription1),amitochondrialproteinthatisreleasedintothecytoplasmupononsetofapoptosis.Ithasbeensuggestedthatthiscomplexturnsoffasurvival-promotinggenetranscriptionprogramcontrolledbytheTLEproteinfamily.Interestingly,apoptosisofcellstransfectedwithAESandBit1couldbeinhibitedifthecellswereallowedtoattachtofibronectinthroughthealpha5beta1integrinsuggestingthattheBit1-AESpathwaycontributingtoanoikisisregulatedbyintegrins,andinparticular,thealpha5beta1integrin.
