Background

Accumulationoftheamyloid-betapeptide(Abeta)inthecerebralcortexisacriticaleventinthepathogenesisofAlzheimersdisease.Thebeta-amyloidproteinprecursor(APP)iscleavedbyoneoftwobeta-secretases(BACEandBACE2),producingasolublederivativeoftheproteinandamembraneanchored99-aminoacidcarboxy-terminalfragment(C99).TheC99fragmentservesassubstrateforgamma-secretasetogeneratethe4kDaamyloid-betapeptide(Abeta),whichisdepositedintheAlzheimersdiseasepatientsbrains.Recently,DeathReceptor6(DR6)wasfoundtointeractwithanamino-terminalfragmentofthebeta-amyloidprotein(N-APP)inneurons,activatingacaspase6-dependentapoptoticeventleadingtoaxonaldegenerationandpruningduringdevelopment,suggestingthatthesetwoproteinsareinvolvedinneuraldevelopmentandmaypossiblyplayaroleinAlzheimersdisease.
